Protestantism is the largest grouping of Christians in the United States, with its combined denominations collectively comprising about 43% of the country's population (or 141 million people) in 2019. Other estimates suggest that 48.5% of the U.S. population (or 157 million people) is Protestant. Simultaneously, this corresponds to around 20% of the world's total Protestant population. The U.S. contains the largest Protestant population of any country in the world. Baptists comprise about one-third of American Protestants. The Southern Baptist Convention is the largest single Protestant denomination in the U.S., comprising one-tenth of American Protestants. Twelve of the original Thirteen Colonies were Protestant, with only Maryland having a sizable Catholic population due to Lord Baltimore's religious tolerance.

The country's history is often traced back to the Pilgrim Fathers whose Brownist beliefs motivated their move from England to the New World. These English Dissenters, who also happened to be Puritans—and therefore Calvinists—, were first to settle in what was to become the Plymouth Colony. America's Calvinist heritage is often underlined by various experts, researchers and authors, prompting some to declare that the United States was "founded on Calvinism", while also underlining its exceptional foundation as a Protestant majority nation. American Protestantism has been diverse from the very beginning with large numbers of early immigrants being Anglican, various Reformed, Lutheran, and Anabaptist. In the next centuries, it diversified even more with the Great Awakenings throughout the country.

In the United States, evangelicalism is a movement among Protestant Christians who believe in the necessity of being born again, emphasize the importance of evangelism, and affirm traditional Protestant teachings on the authority as well as the historicity of the Bible. Comprising nearly a quarter of the U.S. population, evangelicals are a diverse group drawn from a variety of backgrounds, including nondenominational churches, Pentecostal, Baptist, Reformed, Methodist, Mennonite, Plymouth Brethren, and Quaker.

Evangelicalism has played an important role in shaping American religion and culture. The First Great Awakening of the 18th century marked the rise of evangelical religion in colonial America. As the revival spread throughout the Thirteen Colonies, evangelicalism united Americans around a common faith. The Second Great Awakening of the early 19th century led to what historian Martin Marty calls the "Evangelical Empire", a period in which evangelicals dominated U.S. cultural institutions, including schools and universities. Evangelicals of this era in the northern United States were strong advocates of reform. They were involved in the temperance movement and supported the abolition of slavery, in addition to working toward education and criminal justice reform. In the southern United States, evangelicals split from their northern counterparts on the issue of slavery, establishing new denominations that opposed abolition and defended the practice of racial slavery upon which the South's expanding cash-crops-for-export agricultural economy was built. During the Civil War, each side confidently preached in support of its own cause using Bible verses and Evangelical arguments, which exposed a deep theological conflict that had been brewing for decades and would continue long after Lee's surrender at Appomattox.

The Whig Party was a mid-19th century political party in the United States. Alongside the Democratic Party, it was one of two major parties from the late 1830s until the early 1850s and part of the Second Party System. As well as four Whig presidents (William Henry Harrison, John Tyler, Zachary Taylor, and Millard Fillmore), other prominent members included Henry Clay, Daniel Webster, Rufus Choate, William Seward, John J. Crittenden, and John Quincy Adams (whose presidency ended prior to the formation of the Whig Party). The Whig base of support was amongst entrepreneurs, professionals, Protestant Christians (particularly Evangelicals), the urban middle class, and nativists.

The party was hostile towards the ideology of "manifest destiny", territorial expansion into Texas and the Southwest, and the Mexican–American War. It disliked presidential power, as exhibited by Andrew Jackson and James K. Polk, and preferred congressional dominance in lawmaking. Members advocated modernization, meritocracy, the rule of law, protections against majority rule, and vigilance against executive tyranny. They favored an economic program known as the American System, which called for a protective tariff, federal subsidies for the construction of infrastructure, and support for a national bank. The party was active in both the Northern and Southern United States and did not take a firm stance on slavery, but Northern Whigs tended to be less supportive than their Democratic counterparts.

The Catholic Charismatic Renewal has been described as a "current of grace". It began in 1967 when Catholics from Duquesne University attended a Protestant worship service and claimed to have been "baptized in the Holy Spirit". It is heavily influenced by American Protestantism, especially Evangelicalism and Pentecostalism, emphasizing a "personal relationship with Jesus", deep emotional experiences, and expressing the "gifts of the Holy Spirit".

The Prohibition era was the period from 1920 to 1933 when the United States prohibited the production, importation, transportation, and sale of alcoholic beverages. The alcohol industry was curtailed by a succession of state legislatures, and Prohibition was formally introduced nationwide under the Eighteenth Amendment to the United States Constitution, ratified on January 16, 1919. Prohibition ended with the ratification of the Twenty-first Amendment, which repealed the Eighteenth Amendment on December 5, 1933.

A wide coalition of mostly Protestants, prohibitionists first attempted to end the trade in alcoholic drinks during the 19th century. They aimed to heal what they saw as an ill society beset by alcohol-related problems such as alcoholism, domestic violence, and saloon-based political corruption. Many communities introduced alcohol bans in the late 19th and early 20th centuries, and enforcement of these new prohibition laws became a topic of debate. Prohibition supporters, called "drys", presented it as a battle for public morals and health. The movement was taken up by progressives in the Prohibition, Democratic, and Republican parties, and gained a national grassroots base through the Woman's Christian Temperance Union. After 1900, it was coordinated by the Anti-Saloon League. Opposition from the beer industry mobilized "wet" supporters from the wealthy Catholic and German Lutheran communities, but the influence of these groups receded from 1917 following the entry of the U.S. into the First World War against Germany.

Christian fundamentalism, also known as fundamental Christianity or fundamentalist Christianity, is a religious movement emphasizing biblical literalism. In its modern form, it began in the late 19th and early 20th centuries among British and American Protestants as a reaction to theological liberalism and cultural modernism. Fundamentalists argued that 19th-century modernist theologians had misunderstood or rejected certain doctrines, especially biblical inerrancy, which they considered the fundamentals of the Christian faith.

Fundamentalists are almost always described as upholding beliefs in biblical infallibility and biblical inerrancy, in keeping with traditional Christian doctrines concerning biblical interpretation, the role of Jesus in the Bible, and the role of the church in society. Fundamentalists usually believe in a core of Christian beliefs, typically called the "Five Fundamentals". These arose from the Presbyterian Church issuance of "The Doctrinal Deliverance of 1910". The mainline Protestants (sometimes also known as oldline Protestants) are a group of Protestant denominations in the United States and Canada largely of the theologically liberal or theologically progressive persuasion that contrast in history and practice with the largely theologically conservative evangelical, fundamentalist, charismatic, confessional, Confessing Movement, historically Black church, and Global South Protestant denominations and congregations. Some make a distinction between "mainline" and "oldline", with the former referring only to denominational ties and the latter referring to church lineage, prestige and influence. However, this distinction has largely been lost to history and the terms are now nearly synonymous.

Mainline Protestant churches have stressed social justice and personal salvation and, both politically and theologically, tend to be more liberal than non-mainline Protestant churches. Mainline Protestant churches share a common approach that often leads to collaboration in organizations such as the National Council of Churches, and because of their involvement with the ecumenical movement, they are sometimes given the alternative label of ecumenical Protestantism (especially outside the United States). While in 1970 the mainline Protestant churches claimed most Protestants and more than 30 percent of the American population as members, as of 2009 they were a minority among American Protestants, claiming approximately 15 percent of American adults. In 2024, approximately 13.1% of Americans were white non-Hispanic mainline Protestants according to the Public Religion Research Institute's Census of American Religion.
