In neuroanatomy, the primary somatosensory cortex is located in the postcentral gyrus of the brain's parietal lobe, and is part of the somatosensory system. It was initially defined from surface stimulation studies of Wilder Penfield, and parallel surface potential studies of Bard, Woolsey, and Marshall. Although initially defined to be roughly the same as Brodmann areas 3, 1 and 2, more recent work by Kaas has suggested that for homogeny with other sensory fields only area 3 should be referred to as "primary somatosensory cortex", as it receives the bulk of the thalamocortical projections from the sensory input fields.

At the primary somatosensory cortex, tactile representation is orderly arranged (in an inverted fashion) from the toe (at the top of the cerebral hemisphere) to mouth (at the bottom). However, some body parts may be controlled by partially overlapping regions of cortex. Each cerebral hemisphere of the primary somatosensory cortex only contains a tactile representation of the opposite (contralateral) side of the body. The amount of primary somatosensory cortex devoted to a body part is not proportional to the absolute size of the body surface, but, instead, to the relative density of cutaneous tactile receptors located on that body part. The density of cutaneous tactile receptors on a body part is generally indicative of the degree of sensitivity of tactile stimulation experienced at said body part. For this reason, the human lips and hands have a larger representation than other body parts.

Interoception is the collection of senses providing information to the organism about the internal state of the body. This can be both conscious and subconscious. It encompasses the brain's process of integrating signals relayed from the body into specific subregions—like the brainstem, thalamus, insula, somatosensory, and anterior cingulate cortex—allowing for a complex and highly accurate representation of the physiological state of the body. This is important for maintaining homeostatic conditions in the body and, potentially, facilitating self-awareness.

Interoceptive signals are projected to the brain via a diversity of neural pathways, in particular from the lamina I of the spinal cord along the spinothalamic pathway and through the projections of the solitary nucleus, that allow for the sensory processing and prediction of internal bodily states. Misrepresentations of internal states, or a disconnect between the body's signals and the brain's interpretation and prediction of those signals, have been suggested to underlie conditions such as anxiety, depression, panic disorder, anorexia nervosa, bulimia nervosa, posttraumatic stress disorder (PTSD), obsessive compulsive disorder (OCD), attention deficit hyperactivity disorder (ADHD), alexithymia, somatic symptom disorder, and illness anxiety disorder.

The sensory cortex can refer sometimes to the primary somatosensory cortex, or it can be used as a term for the primary and secondary cortices of the different senses (two cortices each, on left and right hemisphere): the visual cortex on the occipital lobes, the auditory cortex on the temporal lobes, the primary olfactory cortex on the uncus of the piriform region of the temporal lobes, the gustatory cortex on the insular lobe (also referred to as the insular cortex), and the primary somatosensory cortex on the anterior parietal lobes. Just posterior to the primary somatosensory cortex lies the somatosensory association cortex or area, which integrates sensory information from the primary somatosensory cortex (temperature, pressure, etc.) to construct an understanding of the object being felt. Inferior to the frontal lobes are found the olfactory bulbs, which receive sensory input from the olfactory nerves and route those signals throughout the brain. Not all olfactory information is routed to the olfactory cortex: some neural fibers are routed to the supraorbital region of the frontal lobe, while others are routed directly to limbic structures. The direct limbic connection makes the olfactory sense unique.

The brain cortical regions are related to the auditory, visual, olfactory, and somatosensory (touch, proprioception) sensations, which are located lateral to the lateral fissure and posterior to the central sulcus, that is, more toward the back of the brain. The cortical region related to gustatory sensation is located anterior to the central sulcus.

In neuroanatomy, the central sulcus (also central fissure, fissure of Rolando, or Rolandic fissure, after Luigi Rolando) is a sulcus, or groove, in the cerebral cortex in the brains of vertebrates. It is sometimes confused with the longitudinal fissure.

It is the location of the primary somatosensory cortex, the main sensory receptive area for the sense of touch. Primary somatosensory cortex in the context of Ventral posterolateral nucleus

The ventral posterolateral nucleus (VPL) is one of the subdivisions of the ventral posterior nucleus in the ventral nuclear group of the thalamus. It relays sensory information from second-order neurons of the neospinothalamic tract and the medial lemniscus (of the dorsal column-medial lemniscus pathway), which synapse with third-order neurons in the nucleus. These then project to the primary somatosensory cortex in the postcentral gyrus.

There is uncertainty regarding the location of VMpo (posterior part of ventral medial nucleus), as determined by spinothalamic tract (STT) terminations and calcium-binding protein staining, and several authorities do not consider its existence as being proved.
