Yersinia pestis (Y. pestis; formerly Pasteurella pestis) is a gram-negative, non-motile, coccobacillus bacterium without spores. It is related to pathogens Yersinia enterocolitica, and Yersinia pseudotuberculosis, from which it evolved. Yersinia pestis is responsible for the disease plague, which caused the Plague of Justinian and the Black Death, one of the deadliest pandemics in recorded history. Plague takes three main forms: pneumonic, septicemic, and bubonic. Y. pestis is a facultative anaerobic parasitic bacterium that can infect humans primarily via its host the Oriental rat flea (Xenopsylla cheopis), but also through aerosols and airborne droplets for its pneumonic form. As a parasite of its host, the rat flea, which is also a parasite of rats, Y. pestis is a hyperparasite.

Y. pestis was discovered in 1894 by Alexandre Yersin, a Swiss/French physician and bacteriologist from the Pasteur Institute, during an epidemic of the plague in Hong Kong. Yersin was a member of the Pasteur school of thought. Kitasato Shibasaburō, a Japanese bacteriologist who practised Koch's methodology, was also engaged at the time in finding the causative agent of the plague. However, Yersin actually linked plague with a bacillus, initially named Pasteurella pestis; it was renamed Yersinia pestis in 1944.

The Black Death was a bubonic plague pandemic that occurred in Europe from 1346 to 1353. It was one of the most fatal pandemics in human history; as many as 50 million people perished, perhaps 50% of Europe's 14th-century population. The disease is caused by the bacterium Yersinia pestis and spread by fleas and through the air. One of the most significant events in European history, the Black Death had far-reaching population, economic, and cultural impacts. It was the beginning of the second plague pandemic. The plague created religious, social and economic upheavals, with profound effects on the course of European history.

The origin of the Black Death is disputed. Genetic analysis suggests Yersinia pestis bacteria evolved approximately 7,000 years ago, at the beginning of the Neolithic, with flea-mediated strains emerging around 3,800 years ago during the late Bronze Age. The immediate territorial origins of the Black Death and its outbreak remain unclear, with some evidence pointing towards Central Asia, China, the Middle East, and Europe. The pandemic was reportedly first introduced to Europe during the siege of the Genoese trading port of Kaffa in Crimea by the Golden Horde army of Jani Beg in 1347. From Crimea, it was most likely carried by fleas living on the black rats that travelled on Genoese ships, spreading through the Mediterranean Basin and reaching North Africa, West Asia, and the rest of Europe via Constantinople, Sicily, and the Italian Peninsula. There is evidence that once it came ashore, the Black Death mainly spread from person-to-person as pneumonic plague, thus explaining the quick inland spread of the epidemic, which was faster than would be expected if the primary vector was rat fleas causing bubonic plague. In 2022, it was discovered that there was a sudden surge of deaths in what is today Kyrgyzstan from the Black Death in the late 1330s; when combined with genetic evidence, this implies that the initial spread may have pre-dated, by nearly two decades, the 14th-century Mongol conquests previously postulated as the cause.

Yersinia pestis in the context of Bubonic plague

Bubonic plague is one of three types of plague caused by the bacterium Yersinia pestis. One to seven days after exposure to the bacteria, flu-like symptoms develop. These symptoms include fever, headaches, and vomiting, as well as swollen and painful lymph nodes occurring in the area closest to where the bacteria entered the skin. Acral necrosis, the dark discoloration of skin, is another symptom. Occasionally, swollen lymph nodes, known as "buboes", may break open.

The three types of plague are the result of the route of infection: bubonic plague, septicemic plague, and pneumonic plague. Bubonic plague is mainly spread by infected fleas from small animals. Yersinia pestis in the context of First plague pandemic

The first plague pandemic was the first historically recorded Old World pandemic of plague, the contagious disease caused by the bacterium Yersinia pestis. Also called the early medieval pandemic, it began with the Plague of Justinian in 541 and continued until 750 or 767. At least fifteen to eighteen major waves of plague following the Justinianic plague have been identified from historical records. The pandemic affected the Mediterranean Basin most severely and most frequently, but also infected the Near East and Northern Europe, and potentially East Asia as well. The Roman emperor Justinian I's name is sometimes applied to the whole series of plague epidemics in late antiquity.

The pandemic is best known from its first and last outbreaks: the Justinianic Plague of 541–549, described by the contemporary Roman historian Procopius, and the late 8th century plague of Naples described by Neapolitan historian John the Deacon in the following century (distinct from the Naples Plague of the 17th century, during the second plague pandemic). Other accounts from contemporaries of the pandemic are included in the texts of Evagrius Scholasticus, John of Ephesus, Gregory of Tours, Paul the Deacon, and Theophanes the Confessor; most seem to have believed plague was a divine punishment for human misdeeds.

Gram-negative bacteria are bacteria that, unlike Gram-positive bacteria, do not retain the crystal violet stain used in the Gram staining method of bacterial differentiation. Their defining characteristic is that their cell envelope consists of a thin peptidoglycan cell wall sandwiched between an inner (cytoplasmic) membrane and an outer membrane. These bacteria are found in all environments that support life on Earth.

Within this category, notable species include the model organism Escherichia coli, along with various pathogenic bacteria, such as Pseudomonas aeruginosa, Chlamydia trachomatis, and Yersinia pestis. They pose significant challenges in the medical field due to their outer membrane, which acts as a protective barrier against numerous antibiotics (including penicillin), detergents that would normally damage the inner cell membrane, and the antimicrobial enzyme lysozyme produced by animals as part of their innate immune system. Yersinia pestis in the context of Non-motile bacteria

Non-motile bacteria are bacteria species that lack the ability and structures that would allow them to propel themselves, under their own power, through their environment. When non-motile bacteria are cultured in a stab tube, they only grow along the stab line. If the bacteria are mobile, the line will appear diffuse and extend into the medium. The cell structures that provide the ability for locomotion are the cilia and flagella. Coliform and Streptococci are examples of non-motile bacteria as are Klebsiella pneumoniae, and Yersinia pestis. Motility is one characteristic used in the identification of bacteria and evidence of possessing structures: peritrichous flagella, polar flagella and/or a combination of both.

Though the lack of motility might be regarded a disadvantage, some non-motile bacteria possess structures that allow their attachment to eukaryotic cells, like GI mucousal cells.
