The Interstate serves the state's second-most populous city, Lexington. Outside of it, the route is mostly rural or suburban in nature, mainly providing access to other cities via state and U.S. Highways. The major landscapes traversed by I-75 include the rolling hills and mountains of the Cumberland Plateau, the flat Bluegrass region, the urban core of Lexington, and the highly urbanized suburbs of Northern Kentucky; it also very briefly crosses through the Eastern Kentucky Coalfield at its southernmost stretch and passes near the Daniel Boone National Forest in London.

Of the six states which I-75 passes through, the segment in Kentucky is the second-shortest, at 191.78 miles (308.64 km) long. I-75 parallels the older U.S. Route 25 (US 25) and U.S. Route 25E (US 25E) corridors for its entire length in Kentucky. The Interstate was part of the Federal-Aid Highway Act of 1956, with a section of it from the Ohio River at Covington to an unknown location north of Richmond being the first segment of the Interstate Highway, opened in 1957. Unfinished portions of the highway were eventually completed in increments, with the very last section being opened in 1969. Due to the rapid growth and high traffic volume in the Lexington metropolitan area and Northern Kentucky, many widening projects and renovations have been undertaken on I-75 since then. The Interstate has one auxiliary route, I-275, a beltway encircling Cincinnati.

Interstate 75 in Kentucky in the context of U.S. Route 25E

U.S. Route 25E (US 25E) is the eastern branch of US 25 from Newport, Tennessee, where US 25 splits into US 25E and US 25W, to North Corbin, Kentucky, where the two highways rejoin. The highway, however, continues as US 25E for roughly two miles (3.2 km) until it joins Interstate 75 (I-75) in the Laurel County community of North Corbin at exit 29. The highway serves the Appalachia regions of Kentucky's Cumberland Plateau and the Ridge-and-Valley section of East Tennessee, including the urbanized areas of Corbin and Middlesboro in Kentucky and Morristown in Tennessee.

US 25E follows the original pathway of early U.S. pioneer Daniel Boone's Wilderness Road, which contributed to the increased settlement of Appalachia given its access through the rugged Cumberland Gap. By 1815, the route became the first state-funded road in Tennessee and, in 1923, unsigned State Route 32 (SR 32) in the Tennessee State Route System in its statewide entirety. It would be recognized as part of the Dixie Highway, one of the earliest federal auto trails, in 1915. The route was officially established as US 25E with the creation of the U.S. Numbered Highway System in 1926.

Corbin is a home rule-class city in Whitley, Knox and Laurel counties in the southeastern portion of the U.S. state of Kentucky. As of the 2020 census, the city population was 7,856.

Corbin is on Interstate 75 and US Route 25W, about halfway between Knoxville, Tennessee, and Lexington, Kentucky.

The Florence Y'all Water Tower is a water tower owned by the city of Florence, Kentucky, United States. It stands between the Florence Mall and Interstate 75/Interstate 71, where it is seen by millions of interstate motorists annually. The tower, built in 1974 and initially painted with the words FLORENCE MALL in giant letters, became a regional landmark after the M was changed to Y' to address legal concerns.

The water tower, which can hold about 1 million US gallons (3,800 m), stands in Boone County in northern Kentucky, due south of Runway 18L/36R of Cincinnati/Northern Kentucky International Airport and between I-75/I-71 and Mall Circle Road, northeast of the mall.
