Denominations have varied conceptions of holy orders. In some Lutheran and Anglican churches the traditional orders of bishop, priest and deacon are bestowed using ordination rites contained within ordinals. The extent to which ordination is considered sacramental in these traditions has, however, been a matter of some internal dispute. Baptists are among the denominations that do not consider ministry as being sacramental in nature and would not think of it in terms of "holy orders" as such. Historically, the word "order" (Latin ordo) designated an established civil body or corporation with a hierarchy, and ordinatio meant legal incorporation into an ordo. The word "holy" refers to the church. In context, therefore, a holy order is set apart for ministry in the church. Other positions, such as pope, patriarch, cardinal, monsignor, archbishop, archimandrite, archpriest, protopresbyter, hieromonk, protodeacon and archdeacon, are not sacramental orders but specialized ministries.

There are also friars outside of the Catholic Church, such as within the Evangelical-Lutheran Churches and Anglican Communion. The term, first used in the 12th or 13th century, distinguishes the mendicants' itinerant apostolic character, exercised broadly under the jurisdiction of a superior general, from the older monastic orders' allegiance to a single monastery formalized by their vow of stability. A friar may be in holy orders or be a non-ordained brother. The most significant orders of friars are the Dominicans, Franciscans, Augustinians, and Carmelites.

The sacrament of holy orders in the Catholic Church includes three orders of men: bishops, priests, and deacons, in decreasing order of rank, collectively comprising the clergy. In the phrase "holy orders", the word "holy" means "set apart for a sacred purpose". The word "order" designates an established civil body or corporation with a hierarchy, and ordination means legal incorporation into an order. In context, therefore, a group with a hierarchical structure that is set apart for ministry in the Church.

Men in the last year of seminary training who are seeking ordination as a priest are typically ordained to the "transitional diaconate", while men who are not seeking priestly ordination are instead ordained to the "permanent diaconate". Deacons, whether transitional or permanent, receive faculties to preach, to perform baptisms, and to witness marriages (either assisting the priest at the Mass, or officiating at a wedding not involving a Mass). They may assist at services where Holy Communion is given, such as the Mass, and they are considered the ordinary dispenser of the Precious Blood (the wine) when Communion is given in both types and a deacon is present, but they may not celebrate the Mass themselves. They may officiate at a funeral service not involving a Mass, including a visitation (wake) or the graveside service at burial.After six months or more as a transitional deacon, a man will be ordained to the priesthood. Priests may preach, perform baptisms, witness marriages, hear confessions and give absolutions, anoint the sick, and celebrate the Eucharist or the Mass. Some priests are later chosen to be bishops, who are the ordinary ministers of confirmation and holy orders; bishops may ordain priests, deacons, and other bishops.

A pastor (abbreviated to "PsPr", "Pstr.", "Ptr." or "Psa" (both singular), or "Ps" (plural)) is the leader of a Christian congregation who also gives advice and counsel to people from the community or congregation. In Lutheranism, Catholicism, Eastern Orthodoxy, Oriental Orthodoxy and Anglicanism, pastors are always ordained. In Methodism, pastors may be either licensed or ordained.

The New Testament typically uses the words "bishops" (Acts 20:28) and "presbyter" (1 Peter 5:1) to indicate the ordained leadership in early Christianity. However, the word "pastor" (ποιμήν/poimēn) is only used one time to describe church leadership in the New Testament in Ephesians 4:11 which says, "that some would be apostles, some prophets, some evangelists, some pastors and teachers" (NRSV). Peter instructs church leadership using the verb, "to tend the flock' (lit. 'to shepherd,' ποιμαίνω/poimainō) of God that is in your charge, exercising the oversight (1 Peter 5:2 NRSV). The words "bishop" and "presbyter" were sometimes used in an interchangeable way, such as in Titus 1:5-6. However, there is ongoing dispute between branches of Christianity over whether there are two ordained classes (presbyters and deacons), or three (bishops, priests, and deacons). The first view is affirmed by some Presbyterians. On the other hand, Christians of the Roman Catholic, Persian, Eastern Orthodox, Oriental Orthodox, Moravian, Scandinavian Lutheran, Anglican, Old Catholic and other Presbyterian traditions maintain the latter view, with all but the Presbyterians affirming the doctrine of apostolic succession.

Holy orders in the context of Catholic priest

The priesthood is the office of the ministers of religion, who have been commissioned ("ordained") with the holy orders of the Catholic Church. Technically, bishops are a priestly order as well; however, in common English usage priest refers only to presbyters and pastors (parish priests). The church's doctrine also sometimes refers to all baptised members (inclusive of the laity) as the "common priesthood", which can be confused with the ministerial priesthood of the ordained clergy.

The church has different rules for priests in the Latin Church–the largest Catholic particular church–and in the 23 Eastern Catholic Churches. Notably, priests in the Latin Church must take a vow of celibacy, whereas most Eastern Catholic Churches permit married men to be ordained. Deacons are male and usually belong to the diocesan clergy, but, unlike almost all Latin Church (Western Catholic) priests and all bishops from Eastern or Western Catholicism, they may marry as laymen before their ordination as clergy. The priesthood is a vocation for men; a similar but distinct vocation for women is the sisterhood. The Catholic Church teaches that when a man participates in priesthood after the Sacrament of Holy Orders, he acts in persona Christi Capitis, representing the person of Christ.

Holy orders in the context of Priesthood of all believers

The priesthood of all believers is the common priesthood of all Christians (a concept broadly accepted by all churches), while the term can also refer to a specific Protestant understanding that this universal priesthood precludes the ministerial priesthood (i.e., holy orders) found in some other churches, including Catholicism and Eastern Orthodoxy.

The inclusionary Catholic version proposes a common priesthood that is different from both holy orders and the priesthood of Christ. The exclusionary version, elaborated in the theology of Martin Luther, Ulrich Zwingli and John Calvin among other reformers, became prominent as a tenet of Protestant Christian doctrine, though the exact meaning of the belief and its implications vary widely among denominations.
