A Petri dish (alternatively known as a Petri plate or cell-culture dish) is a shallow transparent lidded dish that biologists use to hold growth medium in which cells can be cultured, originally, cells of bacteria, fungi, and small mosses. The container is named after its inventor, German bacteriologist Julius Richard Petri. It is the most common type of culture plate. The Petri dish is one of the most common items in biology laboratories and has entered popular culture. The term is sometimes written in lower case, especially in non-technical literature.

What was later called Petri dish was originally developed by German physician Robert Koch in his private laboratory in 1881, as a precursor method. Petri, as assistant to Koch, at Berlin University made the final modifications in 1887 as used today.

Yersinia pestis (Y. pestis; formerly Pasteurella pestis) is a gram-negative, non-motile, coccobacillus bacterium without spores. It is related to pathogens Yersinia enterocolitica, and Yersinia pseudotuberculosis, from which it evolved. Yersinia pestis is responsible for the disease plague, which caused the Plague of Justinian and the Black Death, one of the deadliest pandemics in recorded history. Plague takes three main forms: pneumonic, septicemic, and bubonic. Y. pestis is a facultative anaerobic parasitic bacterium that can infect humans primarily via its host the Oriental rat flea (Xenopsylla cheopis), but also through aerosols and airborne droplets for its pneumonic form. As a parasite of its host, the rat flea, which is also a parasite of rats, Y. pestis is a hyperparasite.

Y. pestis was discovered in 1894 by Alexandre Yersin, a Swiss/French physician and bacteriologist from the Pasteur Institute, during an epidemic of the plague in Hong Kong. Yersin was a member of the Pasteur school of thought. Kitasato Shibasaburō, a Japanese bacteriologist who practised Koch's methodology, was also engaged at the time in finding the causative agent of the plague. However, Yersin actually linked plague with a bacillus, initially named Pasteurella pestis; it was renamed Yersinia pestis in 1944.

Alexandre Émile John Yersin (22 September 1863 – 1 March 1943) was a Swiss-French physician and bacteriologist. He is remembered for his work as a pioneer in microbiology and immunology. Yersin is the co-discoverer of both the Diphtheria and Tetanus toxins (1890 with Émile Roux) and of the bacillus responsible for the bubonic plague or pest (1894, with Kitasato Shibasaburō). The bacteria was later named in his honour: Yersinia pestis. Yersin also demonstrated for the first time that the same bacillus was present in the rodent as well as in the human disease, thus underlining the possible means of transmission.

Baron Kitasato Shibasaburō (北里 柴三郎; January 29 [O.S. 17 January], 1853 – June 13, 1931) was a Japanese physician and bacteriologist. He is remembered as the co-discoverer of the infectious agent of bubonic plague in Hong Kong during an outbreak in 1894, almost simultaneously with Alexandre Yersin.

Kitasato was nominated for the first annual Nobel Prize in Physiology or Medicine in 1901. Kitasato and Emil von Behring, working together in Berlin in 1890, announced the discovery of diphtheria antitoxin serum. Von Behring was awarded the 1901 Nobel Prize because of this work, but Kitasato was not.

Corynebacterium diphtheriae is a Gram-positive pathogenic bacterium that causes diphtheria. It is also known as the Klebs–Löffler bacillus because it was discovered in 1884 by German bacteriologists Edwin Klebs (1834–1913) and Friedrich Löffler (1852–1915). These bacteria are usually harmless, unless they are infected by a bacteriophage carrying a gene which gives rise to a toxin. This toxin causes the disease. Diphtheria is caused by the adhesion and infiltration of the bacteria into the mucosal layers of the body, primarily affecting the respiratory tract and causing the subsequent release of an exotoxin. The toxin has a localized effect on skin lesions, as well as a metastatic, proteolytic effects on other organ systems in severe infections. Originally a major cause of childhood mortality, diphtheria has been almost entirely eradicated due to the vigorous administration of the diphtheria vaccination in the 1910s.

Diphtheria is no longer transmitted as frequently due to the development of the vaccine, DTaP. Although diphtheria outbreaks continue to occur, this is often in developing countries where the majority of the population is not vaccinated.

Gram stain (Gram staining or Gram's method) is a method of staining used to classify bacterial species into two large groups: gram-positive bacteria and gram-negative bacteria. It may also be used to diagnose a fungal infection. The name comes from the Danish bacteriologist Hans Christian Gram, who developed the technique in 1884.

Gram staining differentiates bacteria by the chemical and physical properties of their cell walls. Gram-positive cells have a thick layer of peptidoglycan in the cell wall that retains the primary stain, crystal violet. Gram-negative cells have a thinner peptidoglycan layer that allows the crystal violet to wash out on addition of ethanol. They are stained pink or red by the counterstain, commonly safranin or fuchsine. Lugol's iodine solution is always added after addition of crystal violet to form a stable complex with crystal violet that strengthens the bonds of the stain with the cell wall.
