The earliest record of the church of Antioch is given in Acts 11, stating that some "men of Cyprus and Cyrene, which, when they had come to Antioch, spoke to the Grecians, preaching the Lord Jesus [...] and a great number believed, and turned to the Lord." Later, at the start of their missionary journeys, Paul the Apostle (also called Saul) and Barnabas preached in Antioch for a year, and followers of the church were called "Christians" for the first time.

Saint Peter (born Shimon bar Yonah; 1 BC – AD 64/68), also known as Peter the Apostle, Simon Peter, Simeon, Simon, or Cephas, was one of the Twelve Apostles of Jesus and one of the first leaders of the early Christian Church. He appears repeatedly and prominently in all four New Testament gospels, as well as the Acts of the Apostles. Catholic and Orthodox tradition treats Peter as the first bishop of Rome – or pope – and also as the first bishop of Antioch. According to Christian tradition, Peter was crucified in Rome under Emperor Nero.

The ancient Christian churches venerate Peter as a major saint and the founder of the Church of Antioch and the Church of Rome, but they differ in their attitudes regarding the authority of his successors. According to Catholic teaching, Jesus promised Peter a special position in the Church. In the New Testament, the name "Simon Peter" is found 19 times. He is the brother of Andrew, and they both were fishermen. The Gospel of Mark, in particular, is traditionally thought to show the influence of Peter's preaching and eyewitness accounts. He is also mentioned as Peter or Cephas in Paul's First Letter to the Corinthians and the Epistle to the Galatians. The New Testament also includes two general epistles, First Peter and Second Peter, which are traditionally attributed to him, but modern scholarship generally rejects the Petrine authorship of both.

The Greek Orthodox Patriarchate of Antioch (Greek: Ελληνορθόδοξο Πατριαρχείο Αντιοχείας), also known as the Antiochian Orthodox Church and legally as the Rūm Orthodox Patriarchate of Antioch and All the East (Arabic: بطريركيّة أنطاكية وسائر المشرق للروم الأرثوذكس, romanizedBaṭriyarkiyyat ʾAnṭākiya wa-Sāʾir al-Mašriq li-r-Rūm al-ʾUrṯūḏuks, lit.'Patriarchate of Antioch and All the East for the Orthodox Rum'), is an autocephalous Greek Orthodox church within the wider communion of Eastern Orthodox Christianity that originates from the historical Church of Antioch. Headed by the Greek Orthodox patriarch of Antioch, it considers itself the successor to the Christian community founded in Antioch by the Apostles Peter and Paul. It is one of the largest Christian denominations of the Middle East, alongside the Copts of Egypt and the Maronites of Lebanon.

Its adherents, known as Antiochian Christians, are a Middle-Eastern semi-ethnoreligious Eastern Christian group residing in the Levant region, including the Hatay Province of Turkey. Many of their descendants now live in the global Eastern Christian diaspora. The number of Antiochian Greek Christians is estimated to be approximately 4.3 million.

The Church of Cyprus (Greek: Ἐκκλησία τῆς Κύπρου, romanizedEkklisia tis Kyprou) is one of the autocephalous Greek Orthodox churches that form part of the communion of the Eastern Orthodox Church. Based in Cyprus, it is one of the oldest Eastern Orthodox autocephalous churches; it claims to have always been independent, although it may have been subject to the Church of Antioch before its autocephaly was recognized in 431 at the Council of Ephesus. The bishop of the ancient capital, Salamis (renamed Constantia by Emperor Constantius II) was constituted metropolitan by Emperor Zeno, with the title archbishop.

The Syriac Orthodox Church (Classical Syriac: ܥܺܕܬܳܐ ܣܽܘܪܝܳܝܬܳܐ ܬܪܺܝܨܰܬ݂ ܫܽܘܒܚܳܐ, romanized: ʿIdto Sūryoyto Trīṣāth Šubḥō), also informally known as the Jacobite Church, is an Oriental Orthodox denomination that originated from the Church of Antioch. The Church currently has around 1.5 million followers worldwide. The Church upholds the Miaphysite doctrine in Christology and employs the Liturgy of Saint James, associated with James the Just. Classical Syriac is the official and liturgical language of the Church.

The supreme head of the Syriac Orthodox Church is the patriarch of Antioch, a bishop who, according to sacred tradition, continues the leadership passed down from Saint Peter. Since 2014, Ignatius Aphrem II has served as the Syriac Orthodox Antiochian patriarch. The Great Church of Antioch was the patriarchal seat and the headquarters of the Church until c. 518, after which Severus of Antioch had to flee to Alexandria, Egypt. After the death of Severus, the patriarchal seat moved from Egypt to different monasteries like the Mor Bar Sauma Monastery; some patriarchs also set up headquarters in Antioch temporarily. Later, Mor Hananyo Monastery was declared as the patriarchal seat and the headquarters of the Church from c. 1160 until 1932. In 1959, the patriarchal seat and headquarters were relocated to the Cathedral of Saint George in Bab Tuma, Damascus, Syria, due to conflicts in the region.

The Patriarch of Antioch is a traditional title held by the bishop of Antioch (modern-day Antakya, Turkey). As the traditional "overseer" (ἐπίσκοπος, episkopos, from which the word bishop is derived) of the first gentile Christian community, the position has been of prime importance in Pauline Christianity from its earliest period. This diocese is one of the few for which the names of its bishops from the apostolic beginnings have been preserved. Today five churches use the title of patriarch of Antioch: one Eastern Orthodox (the Greek Orthodox Church of Antioch); one Oriental Orthodox (the Syriac Orthodox Church); and three Eastern Catholic (the Maronite, Syriac Catholic, and Melkite Greek Catholic Churches).

According to the pre-congregation church tradition, this ancient patriarchate was founded by the Apostle Saint Peter. The patriarchal succession was disputed at the time of the Meletian schism in 362 and again after the Council of Chalcedon in 451, when there were rival Melkite and non-Chalcedonian claimants to the see. Following a 7th-century succession dispute in the Melkite Church, the Maronites also began appointing a Maronite patriarch. After the First Crusade, the Catholic Church began appointing a Latin Church patriarch of Antioch, though this became strictly titular after the Fall of Antioch in 1268, and was abolished completely in 1964. In the 18th century, succession disputes in the Greek Orthodox and Syriac Orthodox Churches of Antioch led to factions of those churches entering into communion with Rome under claimants to the patriarchate: respectively the Melkite Greek Catholic patriarch of Antioch and the Syriac Catholic patriarch of Antioch. Their respective Orthodox progenitors are the Greek Orthodox patriarch of Antioch and the Syriac Orthodox patriarch of Antioch.

Church of Antioch in the context of Liturgy of Saint James

The Liturgy of Saint James is a form of Christian liturgy used by some Eastern Christians of the Byzantine rite and West Syriac Rite. It is developed from an ancient Egyptian form of the Basilean anaphoric family, and is influenced by the traditions of the rite of the Church of Jerusalem, as the Mystagogic Catecheses of Cyril of Jerusalem imply. It became the primary Divine Liturgy in the Church of Antioch and Church of Jerusalem in the early fifth century, soon becoming supplanted by the liturgies of Saint Basil and Saint John Chrysostom. It is still the principal liturgy of the Syriac Orthodox Church, the Malankara Orthodox Syrian Church, the Maronite Church, the Syriac Catholic Church, Syro-Malankara Catholic Church and other churches employing the West Syriac Rite. It is also occasionally used in the Eastern Orthodox Church and Melkite Catholic Church. The Malankara Mar Thoma Syrian Church uses a reformed variant of this liturgy, omitting intercession of saints and prayer for the dead.

The liturgy is attributed with the name of James the Just and patriarch among the Jewish Christians at Jerusalem.
