⭐ Core Definition: Antibody

An antibody (Ab), or immunoglobulin (Ig), is a large protein belonging to the immunoglobulin superfamily which is used by the immune system to identify and neutralize antigens such as bacteria and viruses, including those that cause disease. Each individual antibody recognizes one or more specific antigens, and antigens (a portmanteau of "antibody generator") of virtually any size and chemical composition can be recognized. Each of the branching chains comprising the "Y" of an antibody contains a paratope (the antigen-binding site) that specifically binds to one particular epitope (a specific part of an antigen bound by the paratope) on an antigen, allowing the two molecules to bind together with precision. Using this mechanism, antibodies can effectively "tag" the antigen (or a microbe or an infected cell bearing such an antigen) for attack by cells of the immune system, or can neutralize it directly (for example, by blocking a part of a virus that is essential for its ability to invade a host cell).

Antibodies may be borne on the surface of an immune cell, as in a B cell receptor, or they may exist freely by being secreted into the extracellular space. The term antibody generally refers to the free (secreted) form, while the term immunoglobulin can refer to either forms. Since they are, broadly speaking, the same protein, the terms are often treated as synonymous.

Antibody in the context of Milk

Milk is a white liquid food produced by the mammary glands of lactating mammals. It is the primary source of nutrition for young mammals (including breastfed human infants) before they are able to digest solid food. Milk contains many nutrients, including calcium and protein, as well as lactose and saturated fat; the enzyme lactase is needed to break down lactose. Immune factors and immune-modulating components in milk contribute to milk immunity. The first milk, which is called colostrum, contains antibodies and immune-modulating components that strengthen the immune system against many diseases.

As an agricultural product, milk is collected from farm animals, mostly cattle, on a dairy. It is used by humans as a drink and as the base ingredient for dairy products. The US CDC recommends that children over the age of 12 months (the minimum age to stop giving breast milk or formula) should have two servings of milk products a day, and more than six billion people worldwide consume milk and milk products. The ability for adult humans to digest milk relies on lactase persistence, so lactose intolerant individuals have trouble digesting lactose.

Antibody in the context of Blood types

A blood type (also known as a blood group) is a classification of blood based on the presence and absence of antibodies and inherited antigenic substances on the surface of red blood cells (RBCs). These antigens may be proteins, carbohydrates, glycoproteins, or glycolipids, depending on the blood group system. Some of these antigens are also present on the surface of other types of cells of various tissues. Several of these red blood cell surface antigens can stem from one allele (or an alternative version of a gene) and collectively form a blood group system.

Blood types are inherited and represent contributions from both parents of an individual. As of June 2025, a total of 48 human blood group systems are recognized by the International Society of Blood Transfusion (ISBT). The two most important blood group systems are ABO and Rh; they determine someone's blood type (A, B, AB, and O, with + or − denoting RhD status) for suitability in blood transfusion.

Antibody in the context of Immunization

Immunization, or immunisation, is the process by which an individual's immune system becomes fortified against an infectious agent (known as the immunogen). When this system is exposed to molecules that are foreign to the body, called non-self, it will orchestrate an immune response, and it will also develop the ability to quickly respond to a subsequent encounter because of immunological memory. This is a function of the adaptive immune system. Therefore, by exposing a human, or an animal, to an immunogen in a controlled way, its body can learn to protect itself: this is called active immunization. The most important elements of the immune system that are improved by immunization are the T cells, B cells, and the antibodies B cells produce. Memory B cells and memory T cells are responsible for a swift response to a second encounter with a foreign molecule. Passive immunization is direct introduction of these elements into the body, instead of production of these elements by the body itself.

Immunization happens in various ways, both in the wild and as done by human efforts in health care. Natural immunity is gained by those organisms whose immune systems succeed in fighting off a previous infection, if the relevant pathogen is one for which immunization is even possible. Natural immunity can have degrees of effectiveness (partial rather than absolute) and may fade over time (within months, years, or decades, depending on the pathogen). In health care, the main technique of artificial induction of immunity is vaccination, which is a major form of prevention of disease, whether by prevention of infection (pathogen fails to mount sufficient reproduction in the host), prevention of severe disease (infection still happens but is not severe), or both. Vaccination against vaccine-preventable diseases is a major relief of disease burden even though it usually cannot eradicate a disease. Vaccines against microorganisms that cause diseases can prepare the body's immune system, thus helping to fight or prevent an infection. The fact that mutations can cause cancer cells to produce proteins or other molecules that are known to the body forms the theoretical basis for therapeutic cancer vaccines. Other molecules can be used for immunization as well, for example in experimental vaccines against nicotine (NicVAX) or the hormone ghrelin in experiments to create an obesity vaccine.

Antibody in the context of Multiple myeloma

Multiple myeloma (MM), also known as plasma cell myeloma and simply myeloma, is a cancer of plasma cells, a type of white blood cell that normally produces antibodies. Often, no symptoms are noticed initially. As it progresses, bone pain, anemia, renal insufficiency, and infections may occur. Complications may include hypercalcemia and amyloidosis.

The cause of multiple myeloma is unknown. Risk factors include obesity, radiation exposure, family history, age and certain chemicals. There is an increased risk of multiple myeloma in certain occupations. This is due to the occupational exposure to aromatic hydrocarbon solvents having a role in causation of multiple myeloma. Multiple myeloma is the result of a multi-step malignant transformation, and almost universally originates from the pre-malignant stage monoclonal gammopathy of undetermined significance (MGUS). As MGUS evolves into MM, another pre-stage of the disease is reached, known as smoldering myeloma (SMM).

Antibody in the context of Jehovah's Witnesses and blood transfusions

Jehovah's Witnesses believe that the Bible prohibits Christians from accepting blood transfusions. Their literature states that, "'abstaining from ... blood' means not accepting blood transfusions and not donating or storing their own blood for transfusion." This interpretation of scripture is unusual and is one of the doctrines for which Jehovah's Witnesses are best known.

Jehovah's Witnesses' literature teaches that their refusal of transfusions of whole blood or its four primary components—red cells, white cells, platelets, and plasma—is a non-negotiable religious stand and that those who respect life as a gift from God do not try to sustain life by taking in blood, even in an emergency. Witnesses are taught that the use of fractions such as albumin, immunoglobulins, and hemophiliac preparations are not absolutely prohibited and are instead a matter of personal choice.

In chemistry, polyvalency (or polyvalence, multivalency) is the property of molecules and larger species, such as antibodies, medical drugs, and even nanoparticles surface-functionalized with ligands, like spherical nucleic acids, that exhibit more than one supramolecular interaction. For the number of chemical bonds of atoms, the term "valence" is used (Fig. 1). For both atoms and larger species, the number of bonds may be specified: divalent species can form two bonds; a trivalent species can form three bonds; and so on.

Species that have polyvalency usually show enhanced or cooperative binding compared to their monovalent counterparts. Nanoparticles with multiple nucleic acid strands on their surfaces (e.g., DNA) can form multiple bonds with one another by DNA hybridization to form hierarchical assemblies, some of which are highly crystalline in nature.

Antibody in the context of Colostrum

Colostrum (from Latin, of unknown origin), also known as foremilk, is the first form of milk produced by the mammary glands of humans and other mammals immediately following delivery of the newborn. Animal colostrum may be called beestings, the traditional word from Old English dialects. Most species will begin to generate colostrum just prior to giving birth. Colostrum contains antibodies to protect the newborn against disease and infection, and immune and growth factors and other bioactives. The bioactives found in colostrum are beneficial for a newborn's health, growth and vitality. Colostrum strengthens a baby's immune system.

At birth, the environment of the newborn mammal shifts from the sterile conditions of the mother's uterus, with a constant nutrient supply via the placenta, to the microbe-rich environment outside, with irregular oral intake of complex milk nutrients through the gastrointestinal tract. This transition puts high demands on the gastrointestinal tract of the neonate, as the gut plays an important part in both the digestive system and the immune system. Colostrum contributes significantly to initial immunological defense as well as to the growth, development, and maturation of the neonate's gastrointestinal tract by providing key nutrients and bioactive factors. Bovine colostrum powder is rich in protein and low in sugar and fat. Bovine colostrum can also be used for nonorganic failure to thrive in children and acute non-steroidal anti-inflammatory drug-induced increase in intestinal permeability in males and can boost a neonate's immunity.

Antibody in the context of Antitoxin

An antitoxin is an antibody with the ability to neutralize a specific toxin. Antitoxins are produced by certain animals, plants, and bacteria in response to toxin exposure. Although they are most effective in neutralizing toxins, they can also kill bacteria and other microorganisms. Antitoxins are made within organisms, and can be injected into other organisms, including humans, to treat an infectious disease. This procedure involves injecting an animal with a safe amount of a particular toxin. The animal's body then makes the antitoxin needed to neutralize the toxin. Later, blood is withdrawn from the animal. When the antitoxin is obtained from the blood, it is purified and injected into a human or other animal, inducing temporary passive immunity. To prevent serum sickness, it is often best to use an antitoxin obtained from the same species (e.g. use human antitoxin to treat humans).

Most antitoxin preparations are prepared from donors with high titers of antibody against the toxin, making them hyperimmune globulins.

Purification in a chemical context is the physical separation of a chemical substance of interest from foreign or contaminating substances. Pure results of a successful purification process are termed isolate. The following list of chemical purification methods should not be considered exhaustive.

  • Affinity purification purifies proteins by retaining them on a column through their affinity to antibodies, enzymes, or receptors that have been immobilised on the column.
  • Filtration is a mechanical method to separate solids from liquids or gases by passing the feed stream through a porous sheet such as a cloth or membrane, which retains the solids and allows the liquid to pass through.
  • Centrifugation is a process that uses an electric motor to spin a vessel of fluid at high speed to make heavier components settle to the bottom of the vessel.
  • Evaporation removes volatile liquids from non-volatile solutes, which cannot be done through filtration due to the small size of the substances.
  • Liquid–liquid extraction removes an impurity or recovers a desired product by dissolving the crude material in a solvent in which other components of the feed material are soluble.
  • Crystallization separates a product from a liquid feed stream, often in extremely pure form, by cooling the feed stream or adding precipitants that lower the solubility of the desired product so that it forms crystals. The pure solid crystals are then separated from the remaining liquor by filtration or centrifugation.
  • Recrystallization: In analytical and synthetic chemistry work, purchased reagents of doubtful purity may be recrystallised, e.g. dissolved in a very pure solvent, and then crystallized, and the crystals recovered, in order to improve and/or verify their purity.
  • Trituration removes highly soluble impurities from usually solid insoluble material by rinsing it with an appropriate solvent.
  • Adsorption removes a soluble impurity from a feed stream by trapping it on the surface of a solid material, such as activated carbon, that forms strong non-covalent chemical bonds with the impurity.
  • Chromatography employs continuous adsorption and desorption on a packed bed of a solid to purify multiple components of a single feed stream. In a laboratory setting, mixture of dissolved materials are typically fed using a solvent into a column packed with an appropriate adsorbent, and due to different affinities for solvent (moving phase) versus adsorbent (stationary phase) the components in the original mixture pass through the column in the moving phase at different rates, which thus allows to selectively collect desired materials out of the initial mixture.
  • Smelting produces metals from raw ore, and involves adding chemicals to the ore and heating it up to the melting point of the metal.
  • Refining is used primarily in the petroleum industry, whereby crude oil is heated and separated into stages according to the condensation points of the various elements.
  • Distillation, widely used in petroleum refining and in purification of ethanol separates volatile liquids on the basis of their relative volatilities. There are several type of distillation: simple distillation, steam distillation etc.
  • Water purification combines a number of methods to produce potable or drinking water.
  • Downstream processing refers to purification of chemicals, pharmaceuticals and food ingredients produced by fermentation or synthesized by plant and animal tissues, for example antibiotics, citric acid, vitamin E, and insulin.
  • Fractionation refers to a purification strategy in which some relatively inefficient purification method is repeatedly applied to isolate the desired substance in progressively greater purity.
  • Electrolysis refers to the breakdown of substances using an electric current. This removes impurities in a substance that an electric current is run through
  • Sublimation is the process of changing of any substance (usually on heating) from a solid to a gas (or from gas to a solid) without passing through liquid phase. In terms of purification - material is heated, often under vacuum, and the vapors of the material are then condensed back to a solid on a cooler surface. The process thus in its essence is similar to distillation, however the material which is condensed on the cooler surface then has to be removed mechanically, thus requiring different laboratory equipment.
  • Bioleaching is the extraction of metals from their ores through the use of living organisms.
